eng Comparison of Hilic and Reversed-Phase Methods on Chemically Bonded Silica Columns The aim of work was focused on the optimization of working conditions, especially on the ratio of acetonitrile/water in the mobile phase and the temperature of separation. No retention was observed in range of 20 - 70 % (v/v) of water on all used columns. Cholesterol column provided suitable conditions for HILIC separation, thus a water rich layer could be formed onto its surface. Peaks of separated phenolic acids on cholesterol column mode were symmetrical in HILIC as well as their widths were sufficiently small. Cholesterol groups, the non polar molecules bonded onto silica surface, also presented an excellent medium for usage in RP mode so separated phenolic acids eluted in order of their increasing hydrophobicity caused by their substituted aryl groups.
