eng Lipidomic profiling of patients with cardiovascular diseases using GC/FID and HPLC/MS The goal of this work is the lipidomic profiling of erythrocytes, plasma and lipoprotein fractions samples to distinguish healthy patients and patients with different types of cardiovascular diseases. Gas chromatography with flame ionization detection (GC/FID) is the standard method used for profiling of fatty acids in a biological material as fatty acid methyl esters (FAME) after their transesterification with sodium methoxide. The length of the column, the type and flow rate of carrier gas and the temperature program are important parameters for the separation of FAME using GC/FID. These conditions were optimized using a model mixture of FAME standards and plasma samples of healthy volunteers. The analysis of lipid classes was performed with ultra high performance liquid chromatography - mass spectrometry (UHPLC/MS) using HILIC based on their polarity. In total, 25 fatty acids in the GC/FID analysis and 6 classes of lipids in the HILIC analysis were detected. gas chromatography;fatty acids;lipids
