eng Immobilization of alkaline phosphatase on magnetic particles Several steps in the process of sample preparation are performed by enzymatic reactions. One of these enzymes is alkaline phosphatase. This enzyme is used for in vitro hydrolysis of phosphoesters bounds on phosphoserine, phosphothreonine and phosphotyrosine. Reaction of free enzyme can help with confirmation of phosphorylation on studied proteins and it is frequently used in phosphoproteomics. When we immobilized enzyme on particles, we can gain some benefits compare to use of free enzyme. Immobilized enzymes can be used for more than one reaction, reaction can be done faster, and enzyme used in reaction is fully removed from reaction mixture and do not contaminate our sample. In our work we concentrated on immobilization of alkaline phosphatase on magnetic particles with various functional groups bind to their surface. We compare quantity of active enzyme bound on various particles in different conditions, and verify usability of the immobilized enzymes compared to their free forms. immobilization; alkaline phosphatase; magnetic particles
