eng Determination of activities of desaturases and elongases in type 2 diabetics SCD-1 (stearoyl-CoA desaturase 1) is a liver enzyme essential for biosynthesis of unsaturated fatty acids from the saturated ones (the introduction of the first cis-double bond between carbons 9 and 10), which are essential for the synthesis of glycerol esters and cholesterol in the body. Other desaturases introduce a double bond into the Δ6 and Δ5 positions. Elongases are part of the microsomal system that prolongs fatty acids chain bound to coenzyme A by 2 carbons. 25 anonymized plasma samples randomly selected type 2 diabetic patients were processed in this study. These samples of biological material were also used for verification of analytical method. Furthermore, there were used 6 healthy blood donors plasma samples as a control group. For purposes of the study, samples were categorized into three groups according to level of glycosidic hemoglobin (Hb). This indicator appears to be most reliable because it is affected by poor conditions (such as non fasting) prior to collection, and highlights the development of blood glucose in the last 4 to 6 weeks. The achieved results allowed us to evaluate the potential correlations between lipidic enzymes activities and other parameters like glycosidic hemoglobin, blood glucose, fatty acids and plasma triglycerides. In addition, we examined the usefulness of indices of activity of enzymes in each class for diagnosis or rather for the assessment of disease status in patients already diagnosed. Classes that appear promising for further investigation by both the above criteria, for 9-desaturase are free fatty acids and triglycerides, for 5-desaturase are esters of cholesterol and triglycerides, for the 6-desaturase all classes are potentially useful and for elongase index interesting classes to capture the disease are phospholipids, diglycerides and free fatty acids. For the disease assessment, elongase is not appropriate because of its value did not show significant difference between moderate and severe patients. diabetes mellitus; lipids; desaturase; elongase; gas chromatography; thin layer chromatography
