eng Identification of carbonic anhydrase I immunodominant epitopes recognized by specific autoantibodies of patients with malignancy after autologous stem cell transplantation High-dose radiotherapy (HDT) with autologous stem cell transplantation (ASCT) is the standard treatment for patients with chemosensitive malignant diseases as Hodgkin´s lymphoma, non-Hodgkin´s lymphoma, multiple myeloma etc. In patients, who developed aplastic anemia type syndrome after the ASCT therapy, spontaneous tumor regression was observed. Blood serum analysis by Western blott has revealed high titer of anti-CA I autoantibodies. CA I (mammalian carbonic anhydrase I, EC 4.2.1.1) is wide spread metalloenzyme localized in cytosol of red plasma cells. It is evidence that anti-tumor activity also operates in autoimmunity against hematopoietic stem cells in acquired aplastic anemia (AA). The main goal of this project: the identification of the immunodominant epitope(s) of CA I. The recognition of these immunodominant epitope(s) has a potential to be applied consequently in clinical practice for therapeutic purposes as peptide-based vaccination to develop specific anti-tumor reactivity of patient´s immune system. carbonic anhydrase; immunodominant epitopes; autoantibodies; autologous stem cell transplantation; high-dose radiotherapy
