eng Stability of oxidative stress and anti-oxidant parameters in human serum In epidemiological research it is very important to test the stability of biomarkers as function of both storage time and temperature. In this study the stability of a number of parameters for the assessment of oxidative stress and the anti-oxidant capacity have been tested at two different temperatures for a period of up to one year. In this report, the results of 2 studies are combined in which serum samples of 16 and 34 volunteers have been used. The activity or potency of a number of biomarkers in serum have been determined at T=0 and at several time points (2 weeks and 1, 3, 6, 9 and 12 months) after storage at -20 oC and -70 oC. As oxidative stress biomarkers we have tested two parameters for hydroperoxides, reactive oxygen metabolites (ROM) and total oxidant status (TOS). For the antioxidant status we have tested four parameters, the total antioxidant status (TAS) both first and second generation, the biological antioxidant potential (BAP) and an enzymatic assay (ImAnOx) and in addition an assay for total thiols in serum (TTP). The results of this stability study will be presented in combination with the quality control parameters of the assays during one year. Not all parameters tested were stable during one year of storage. Whether this is due to kit stability or assay performance will be discussed. Stability; oxidative stress; anti-oxidant parameters;
