eng Optimisation of Headspace Solid-Phase Microextraction Method for Identification of Volatile Compounds in Sulphur-Containing Vegetables The aim of this work was comparison of volatile constituents in various kinds of vegetable containing sulphur compounds. A headspace–solid phase microextraction (HS-SPME) was employed as the extraction method. The optimisation of the extraction conditions was performed on chive, and it was based on the use of three SPME fibers with different solid phases: polyacrylate (PA), carboxen/polydimethylsiloxane (CAR/PDMS) and divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS). Each of fibers underwent sequence of extractions based on central composite design (CCD). Four factors of extraction were optimised: temperature and time of the extraction, a volume of saline solution, and a concentration of sodium chloride in saline solution. Fifteen real samples were examined under the optimised extraction conditions (DVB/CAR/PDMS fiber, 70 °C, 50 min, and without saline solution) with a preincubation step of 20 min. Separation and detection of volatile compounds were carried out by gas chromatography-mass spectrometry (GC-MS), when individual peaks (constituents) were compared to mass spectrum libraries and literature.
