eng The introduction of the HPLC method for determination of selected amino acids in human embryo culture medium Despite of the increasing success in infertility treatment methods of assisted reproduction, it still remains a problem to select the best quality embryo that would have a potential for further development. At the present time, embryo selection is based mainly on morphological criteria. Because this approach is very subjective, there is the effort to find more objective method. Amino acids (AA) exogenously supplied into culture medium may have an effect on human pre-implantation embryo development in vitro. The aim of this study was to develop a high-performance liquid chromatography (HPLC) method for assessing changes in levels of selected AA in the human embryo culture medium before and after incubation. Two types of culture medium samples were used – blank culture medium that refers to the medium before incubation and culture medium after incubation of the only one embryo. Filtered medium samples were derivatized with ortho-phthalaldehyde to form highly fluorescent amino acid derivatives that were separated on a reversed-phase column. Simple and selective HPLC method with fluorescence detection for the determination of selected AA in human embryo culture medium has been developed, evaluated and validated. This method is non-invasive and sensitive enough to monitor changes in concentration of selected AA in human embryo culture medium. Results indicate that embryo development can be associated with amino acids turnover.
