eng Isolation and determination of class I hydrophobins Hydrophobins are primary metabolites of microscopic filamentous fungi. These small proteins with molecular weight mostly between 7 and 15 kDa are characterized by pattern of eight Cys residues in their primary structure, which are linked by four disulfide bonds. Class I hydrophobins (e.g. SC3 from Schizophyllum commune) form highly insoluble aggregates, which can only be dissolved with strong acids. The isolation of class I hydrophobins is due to their properties very difficult. In this work, new isolation technique based on high surface activity of hydrophobins to the teflon as a adsorption agent was developed. The selective isolation of hydrophobin SC3 from model mixture of four and ten proteins was achieved using teflon microparticles. The separation of proteins was done using tricine SDS-PAGE electrophoresis and identification was performed using mass spectrometry with matrix-assisted laser desorption ionization (MALDI) connected with orbitrap or TOF as analyzers. hydrophobins, protein analysis, mass spektrometry, electrophoresis
