eng Simple micro-sized amperometric detector integrated into the monolithic capillary column Simple amperometric detector embedded into the monolithic capillary column was designed using carbon working and silver pseudoreference electrodes. It was proven that such two-electrode setup is suitable for on-column detection of dopamine in mixed solvent containing 0.1 M phosphate buffer pH 7 and acetonitrile. Attempts were undertaken to prepare pseudoreference electrode made of carbon fiber coated with metallic silver. Conditions of silver electrodeposition, namely the way of carbon fiber activation, deposition time, and composition of plating solution, were evaluated with respect to uniform coverage and thickness of silver film. The final arrangement of detector utilized silver microwire as reference, which possessed better potential and chemical stability in time comparing to silver-coated carbon fiber. Epoxy sized and unsized carbon fibers were tested as working electrodes in direct amperometry of dopamine. The effect of electrochemical pretreatment was also studied. Influence of detection potential and flow rate on the oxidation signal of dopamine was examined. The dependence of peak current vs. concentration of analyte was linear within the concentration range 5×10−4 – 1×10−7 M dopamine using unsized carbon fibers pretreated in perchloric acid, detection potential of 1 V, and flow rate 10 μL min−1.
