In this work, two HPLC systems were used for the analysis of selected fat-soluble vitamins (e.g. retinol, several tocopherols). Specifically, HPLC-UV system and HPLC system equipped with multichannel coulometric array detector were used. Reversed phase chromatography was chosen in both cases. Column of type C18 as stationary phase, and methanol with water as mobile phase in isocratic elution were used for separation of vitamins using HPLC-UV system. Column of the same type, but more complex mobile phase consisted of a mixture of water, sodium chloride, perchloric acid, acetonitrile and propanol in gradient elution were used for analysis using HPLC with electrochemical detection. Optimized chromatographic conditions for analysis of selected lipophilic vitamins were applied for the determination of these analytes in real samples such as various oils, milk or margarine.
