eng The preparation of hyaluronic acid nanoparticles Hyaluronic acid (HA) is a naturally occurring long linear polysaccharide known for its biocompatibility, biodegradability and non-immunogenicity. We prepared hyaluronic acid nanoparticles (HA-NPs) by cross-linking of the linear polymer HA molecule. The product (HA-NPs) was then measured by dynamic light scattering (DLS). But thanks to the broad size distribution of nanoparticles prepared by this already published method, it was necessary to determine which condition of the preparation corresponds with the final size distribution. For these reasons, the following conditions were altered and studied – the incubation time and the number of steps in the final acetone addition, the amount of adipic acid dihydrazide, and the speed of mixing during incubation. From DLS measurements, we obtained the values of mean size, zeta average, and polydispersity index (PI). The type of motion which seems to be the most convenient for this preparation is the medium speed rotation. Two additions of acetone with each incubation time 10 minutes corresponds with the size around 120 nm. For nanoparticles with the size around 100 nm, the best are two additions of acetone with incubation 20 minutes. And for nanoparticles with size around 80 nm use the addition of acetone three times with incubation 30 minutes. To conclude, this study describes a method of preparation of HA-NPs, which can be modified to achieve nanoparticles of certain size of particles with narrow size distribution. hyaluronic acid; nanoparticles; preparation; optimization; narrow size distribution; DLS
