eng Is it correct to leave out the fluorimetric methods for glutathione assay? Glutathione is the main intracellular antioxidant, which exists in two forms, the reduced and the oxidized. Changes in glutathione concentration serve as an important marker of the redox state inside the cells. Various methods can be used to measure glutathione concentration in biological material, namely enzymatic, fluorimetric or HPLC. Nevertheless, due to the high sensitivity and specificity HPLC methods are preferred to analyze glutathione concentration, in spite of high price and time demand. The aim of our study was to establish analytical parameters of the spectrofluorimetric method, which should be suitable for the analysis of huge amount of samples. This method is based on the well-known principle - reaction between glutathione and o phthaldialdehyde with subsequent production of strongly fluorescent isoindole-like product. To confirm selectivity of this method, we have analysed glutathione concentration in hepatocyte lysates using HPLC method with spectrofluorimetric detection. Hepatocytes are appropriate cells to confirm specificity because of high intracellular level of thiols, which may interfere with the measurement of glutathione. We have proved the specificity of the spectrofluorimetric method and found following analytical parameters: very good sensitivity (detection limit c = 2,5 M; linearity 5 ? 250 M), large accuracy (calibration curve R2 = 0,999) and reproducibility (intra assay CV% = 2,5%, n = 10).
