eng The analysis of tumor samples using shotgun mass spectrometry and novel Lipidomics deals with the understanding of lipid functions in biological systems and progress of many diseases associated with the disruption of lipid metabolism such as atherosclerosis, cancer, obesity, cardiovascular disease, etc. Mass spectrometry coupled with the direct infusion (shotgun approach) or liquid chromatography (LC/MS) is the most frequently used technique in the lipid analysis due to high sensitivity and selectivity. Shotgun approach enables rapid analysis using product ion, neutral loss and the selected reaction monitoring scans typical for triple quadrupoles and related hybrid tandem mass analyzers. The quantitative analysis in shotgun approach is based on the use of at least one internal standard per each lipid class, which is not present in studied biological samples. Shotgun lipidomics was used for the characterization of differences in the lipid composition of lung tumor tissues and surrounding normal tissues followed by the multivariate data analysis.
