eng Automated dual comprehensive HILIC×RP RP×RP liquid chromatography for separation phenolic compounds We investigated possibilities of on-line combining non-correlated two-dimensional systems including a single zwitterionic polymethacrylate monolithic column operated both in the HILIC and in the reversed-phase (RP) modes, in combination with silica monolithic or core-shell alkyl bonded short columns. A monolithic zwitterionic polymethacrylate micro-column prepared in our laboratory shows dual retention mechanism, HILIC and RP. The retention on the micro-column in acetonitrile-rich mobile phase (HILIC) is almost perfectly orthogonal to the retention in the RP mode in more aqueous mobile phases. This micro-column can be used in the first dimension of two-dimensional LC in alternating reversed-phase and HILIC modes, for HILIC×RP and RP×RP comprehensive two-dimensional separations. The fast repeating reversed-phase gradients on a short silica-monolithic or core-shell column in the second dimension can be optimized independently of the actual running first-dimension gradient program to achieve the best resolution and best coverage of separation space. The instrumental setup essentially represents directly coupled two different on-line two-dimensional systems: combining two subsequent complementary on-line 2D (HILIC×RP and RP×RP) separations. The novel system allows obtaining three-dimensional data in a relatively short time with improved total peak capacity with respect to either HILIC×RP or RP×RP two-dimensional separations and gives us more information about the sample.
