eng Pneumatically controlled micro-milled microfluidic device The purpose of the presented microfluidic device is isolation and treatment of glycoprotein samples. Small narrowing in the channel allowed capture and stacking of injected microbeads and formation of packed affinity column. Mixing of the eluted captured glycoproteins with the treating agent was realized inside repeating rectangular pattern in the last portion of the microchannel. Integration of pneumatic valves connected to a computer-controlled multichannel CDA-switchboard enables full automation of the whole sample treatment procedure. The developed device was fabricated using a CNC router. Channels milled in transparent plastic using flat-end mill had rectangular 400 µm wide profile. The channel depth was 250 µm along most of its length but doubled at output branches to reduce back-pressure. Channels were sealed by a PDMS membrane which was sandwiched by another piece of plastic. This piece contained connections for screw-in pneumatic valves managing reagent input and output and flow control. Application of air pressure on the valves pushed a piston into the membrane which sealed the channel. These valves were also constructed from components fabricated on the CNC router. To enable fluidic connections, threaded tapered holes precisely fitted for the 2-56 UNC PEEK ferules were milled through the plastic. CNC router was also implemented in the construction of a fluid dispenser. Device contained a rack of tightly sealed constantly pressurized micro-centrifuge tubes serving as fluid reservoirs or as sample collectors if required. Wider-ID fused silica capillaries were utilized to connect the microfluidic device to the fluid dispenser. Micro-milling using CNC machines is a popular fabrication technique for creating casting masters or microfluidic devices with wider channels.
