eng DETERMINATION OF OXYLIPINS IN HUMAN PLASMA SAMPLES BY UHPLC/MS Oxylipins (eicosanoids, octadecanoids, and docosanoids) are a special family of bioactive lipids generated from polyunsaturated fatty acids (PUFA), such as arachidonic acid, docosahexaenoic acid, or eicosapentaenoic acid. Oxylipins can be formed by either enzymatic pathways using cyclooxygenase (COX), lipoxygenase (LOX), and cytochrome P450 (CYP450) enzyme or non-enzymatic pathways. Oxylipins are connected with many physiological functions, such as the regulation of blood pressure, cell proliferation, or inflammation and with diseases (e.g. cancer, diabetes, and heart diseases). UHPLC/MS method is the most useful for their determination in human plasma samples, where oxylipins are detected by selected reaction monitoring (SRM) and ionized by electrospray ionization in negative-ion mode due to the presence of carboxylic group in their structures. We developed the UHPLC/MS method for determination of 63 oxylipins in human plasma, where we used a mixture of 14 deuterated internal standards for their quantitation. This method was fully validated, applied to 40 human plasma samples (20 healthy volunteers and 20 breast cancer patient), and statistically evaluated using multivariate data analysis (MDA), such as supervised orthogonal partial least square-discriminant analysis (OPLS-DA).
