eng Lipidomic analysis of human plasma and exosomes by various MS techniques Lipidomics is a branch of metabolomics aimed at the analysis of lipid species. Lipids play important roles in cellular functions, and they are potential biomarkers for some human diseases. Nowadays, mass spectrometry (MS) is a key technique for lipids analysis. Ultrahigh-performance supercritical fluid chromatography with electrospray ionization mass spectrometry (UPHSFC/ESI-MS), matrix-assisted laser desorption/ionization coupled with Orbitrap mass spectrometry (MALDI-Orbitrap-MS), and direct infusion mass spectrometry (known as shotgun MS) were used for the analysis of human plasma and exosomes. Exosomes were isolated by ultracetrifugation method, and the total lipid extract was prepared by Folch procedure. Samples of kidney cancer patients and healthy controls were used for the comparison. Goals were lipidomic comparison biological samples and their multivariate data analysis. For lipid quantification, mixture of 16 internal stand was used. Signal stability was controlled by quality control (sample, which was the pooled sample containing internal standards). Data were processed by Water´s tools for the data size reduction and obtained accurate masses. Our laboratory-made software LipidQuant was used for the semi-automated identification, isotopic correction, and quantitation of lipid species. Software Simca was used for scaling and transformation date and for unsupervised (PCA) and supervised modeling (OPLS-DA). The models showed differences between patients and healthy control samples.
