eng Unusual Setup in Comprehensive Two-dimensional Liquid Chromatography Allowing Manipulation with Fractions With the comprehensive two-dimensional liquid chromatography, hundreds of compounds can be analyzed and quantified in a single analysis. The dilution of the sample components, which occurs during their migration throughout the two-dimensional system, increases detection limits of such methods in comparison to the uni-dimensional LC. The excessive band broadening due to the fraction transfer process should be minimized, or the interface with focusing capabilities can be applied, which can utilize trapping columns instead of sampling loops [1]. In the present work, we have developed setup for comprehensive two-dimensional liquid chromatography, allowing us to manipulate with the sample during the collection of the fraction and prior to its analysis in the second separation dimension. The effluent from the first dimension column is collected within the interface and it is available for solvent-replacement, derivatization, or, the sample zone can be pre-concentrated and consecutively transferred into the second dimension with a narrower volume. We have compared our approach with the most commonly used valvetype interface with two sampling loops and with stationary-phase-assisted modulation approach [2]. We have tested the setup using mixtures of natural porphyrin-type colorants and polyphenolic antioxidants. The developed method was applied to the analysis of samples of plant extracts and food products (beverages, confectionery).
