eng Determination of lipids in human plasma samples using RP-UHPLC/MS Lipidomics is a subgroup of metabolomics which study all lipids, their structures, biology, and functions in cells. It aims to understand the function of lipids in biological systems and the mechanism of origin because disorders in lipid metabolism can lead to diabetes, cancer, cardiovascular diseases, obesity, neurodegenerative disorders, and other serious diseases. Lipid analyses are based on mass spectrometry (MS) either with direct infusion (shotgun) or with connection to prior separation. Liquid chromatography is the most common separation technique for lipid analysis which can be easily combined with electrospray ionization MS. Reverse-phase ultrahigh performance liquid chromatography (RP-UHPLC) method separates lipids into lipid species, according to the length of the fatty acyl chains and the number and position of the bonds. The goal of this work is to develop RP-UHPLC/MS method for determination and then subsequent quantitation of lipids in biological samples such as human plasma. RP-UHPLC/MS method using sub-2-µm particles column and the system was coupled to high-resolution mass spectrometry using quadrupole – time of flight (QTOF) mass analyzer. Electrospray ionization (ESI) full scan and MS/MS spectra were measured in both polarity modes with the mass accuracy better than 5 ppm. The identification of individual lipids was based on retention of behavioural characteristics. In total, 478 lipids within 9 polar and nonpolar lipid classes were identified in human plasma samples.
