Proinflammatory M1 macrophages are activated by IFN-g or lipopolysaccharide. M2 macrophages are driven by Th2 or IL-4/TGF-β acting in wound healing and tissue repair. The aim of our study was to compare the oxygen consumption rates in naive M0 macrophages to activated M1 or M2. After activation with IFN-g/LPS or IL-4/TGF-β, the macrophages were assayed by a high-resolution respirometer. Four respiratory states were measured: ROUTINE (at the physiological conditions), LEAK (after inhibition of ATP synthase by oligomycin), ETS (maximum O2 consumption rate after uncoupling), and ROX (residual oxygen consumption). We found no differences at ROUTINE/LEAK/ETS states between M0 and M2 macrophages. On the other hand, M1 macrophages differed from both M0/M2 in all respiratory states. Surprisingly, M1 macrophages did not respond to treatment with mitochondrial inhibitors. The study was supported by grant 23-06536S of the Czech Science Foundation.
