eng USE OF INDOXYL ACETATE AS A SUBSTRATE FOR ACETYLCHOLINESTERASE Acetylcholinesterase (AChE) is a serine hydrolase, that terminates neurotransmission by degrading the neurotransmitter acetylcholine. Increased AChE activity in the cholinergic nervous system can cause signal transduction disorders and is probably one of the causes of the development of Alzheimer's disease. The activity of AChE is usually determined by the Ellman´s method. This method is based on the use of acetylthiocholine as a substrate and 5,5´ dithiobis(2-nitrobenzoic acid) as a chromogen. In this work indoxyl acetate was used as a substrate. Its hydrolysis catalysed by AChE provides a product having an absorption maximum at 670 nm. The aim of this work was to find suitable reaction conditions for spectrophotometric determination of AChE activity using indoxyl acetate as a substrate. Different substrate concentrations and amounts of the enzyme in the reaction mixture were tested. Compared to the Ellman's method, the use of indoxyl acetate as a substrate for AChE has several advantages: there is no need to add chromogen to the reaction mixture and there is no inhibition by the substrate even at higher concentrations.
