eng Utilization of the PCR method for detection of Arcobacter The objective of this study was to accelerate the detection of Arcobacter spp. isolated from foodstuffs and water and observes an influence of samples´ matrix. Method of optimized multiplex PCR was verified on reference strains Arcobacter butzleri CCUG 30484, Arcobacter cryaerophilus CCM 3933 and Arcobacter cryaerophilus CCM 3934. Direct analysis of enrichment broths takeing-off a culture step was performed. Four enrichment media were tested for the acceleration of arcobacters´ detection. F6 selective broth yielded the best results. Poultry, porcine and bovine samples were artificially contaminated and the influnce of samples´ matrix was investigated. A choice combination of an enrichment broth and a plating agar was highly selective. Sorts of meat differ in a content of fat, which had an influence on a detection of arcobacters. 69 samples of food products and water were examined using multiplex PCR. Arcobacter spp. were found in 29 cases (42 %). Positive isolates were confirmed as A. butzleri (24,6 %) and A. cryaerophilus (17,4 %).
